Next-generation sequencing (NGS) is a powerful and cost-effective technology for rapidly determining the DNA or RNA sequence of large numbers of molecules simultaneously. It has transformed genomics by enabling high-throughput sequencing, making it valuable for applications like disease genetics, gene expression analysis, and environmental studies. NGS relies on advanced bioinformatics and various platforms such as Illumina, Ion Torrent, and PacBio. This brief presentation will provide an overview and application of exome sequencing, as well as an analysis workflow that includes quality control, data alignment, variant identification, variant annotation, and prioritization.

Monoclonal gammopathies are a group of disorders involving the abnormal growth of a single type of plasma cell in the bone marrow, leading to the production of monoclonal proteins (M-proteins). These conditions include multiple myeloma, MGUS, and Waldenström macroglobulinemia. Multiple myeloma is the most severe and can cause various health issues, while MGUS is a precursor condition with a lower risk of progression. Diagnosis involves blood tests and bone marrow biopsy, and treatment options range from chemotherapy to observation, depending on the specific disorder. Monitoring is crucial to detect any disease progression.
